Protection of cattle against foot-and-mouth disease by a synthetic peptide   总被引:42,自引:0,他引:42  
A chemically synthesized peptide consisting essentially of two separate regions (residues 141 to 158 and 200 to 213) of a virus coat protein (VP1) from the O1 Kaufbeuren strain of foot-and-mouth disease virus was prepared free of any carrier protein. It elicited high levels of neutralizing antibody and protected cattle against intradermolingual challenge by inoculation with infectious virus. Comparative evaluation of this peptide with a single-site peptide (residues 141 to 158) in guinea pigs suggests the importance of the VP1 carboxyl terminal residues in enhancing the protective response.  相似文献

The epidermal growth factor receptor (EGFR) kinase inhibitors gefitinib and erlotinib are effective treatments for lung cancers with EGFR activating mutations, but these tumors invariably develop drug resistance. Here, we describe a gefitinib-sensitive lung cancer cell line that developed resistance to gefitinib as a result of focal amplification of the MET proto-oncogene. inhibition of MET signaling in these cells restored their sensitivity to gefitinib. MET amplification was detected in 4 of 18 (22%) lung cancer specimens that had developed resistance to gefitinib or erlotinib. We find that amplification of MET causes gefitinib resistance by driving ERBB3 (HER3)-dependent activation of PI3K, a pathway thought to be specific to EGFR/ERBB family receptors. Thus, we propose that MET amplification may promote drug resistance in other ERBB-driven cancers as well.  相似文献

Shorebirds are declining worldwide due to loss and degradation of critical breeding and wintering habitats. Some human-modified habitats, particularly salt-pans which are used by shorebirds in many regions of the world, may help substitute for natural habitats lost for a wide range of species during migration. We studied the influence of landscape characteristics on species richness, abundance, and diversity of shorebirds at 20 sites covering most of the Inner Gulf of Thailand, a landscape with a long history of salt farming. Sites with salt-pans present held significantly higher species richness, abundance and diversity of shorebirds. Areas with larger proportions given over to aquaculture tended to have lower species richness, abundance and diversity. Generalized additive models indicated that landscapes with a larger proportion of tidal flats in conjunction with salt-pans were the best predictors of sites with higher species richness, abundance and diversity. Landscape configurations with higher richness, abundance and diversity of shorebirds also tended to be less fragmented and contained slightly larger patches. Shorebirds appeared to use ponds with exposed mud in salt-pans as both roosting sites and supplementary feeding grounds during high tide. Traditional salt-pans therefore proved to contribute significantly to maintenance of overwintering shorebird populations in this landscape and should be investigated elsewhere in Asian coastal zones. Collaboration between researchers, salt farmers and planning authorities as to how best to maintain salt-pans as potential shorebird roost sites such as in the Inner Gulf of Thailand is urgently needed in order to maintain habitat for shorebird populations in critical wintering and staging areas of this flyway.  相似文献

The anticoccidial activity of maduramicin ammonium (5 mg/kg food) administered alone or with roxarsone (50 mg/kg food) and/or avoparcin (10 mg/kg food) was evaluated in battery-reared broilers artificially challenged with recent field culture mixtures containing Eimeria acervulina, E. maxima and E. tenella or E. mivati, E. necatrix and E. brunetti. 2. Maduramicin ammonium exhibited a high degree of anticoccidial activity and the addition of roxarsone and/or avoparcin in food at recommended concentrations did not adversely affect the activity.  相似文献

Plastic-film-type antitranspirants are more permeable to water vapor than to carbon dioxide. Photosynthesis in treated plants is reduced to the same or greater extent than transpiration, except under conditions of stress, when photosynthesis in treated plants is greater than in controls. This exception is attributed to reduction of stress, as the selective permeability of the films to gases would tend to produce the reverse effect.  相似文献

The Drought Calculator (DC), a spreadsheet-based decision support tool, was developed to help ranchers and range managers predict reductions in forage production due to drought. Forage growth potential (FGP), the fraction of historical average production, is predicted as a weighted average of monthly precipitation from January through June. We calibrated and evaluated the DC tool in the Great Plains of the United States, using FGP and precipitation data from Colorado (CO), North Dakota (ND), and Wyoming (WY). In CO, FGP was most sensitive to precipitation in April and May, in ND to precipitation in April and June, and in WY to precipitation in April, May, and June. Weights in these months ranged from 0.16 to 0.52. Prediction was better for CO and WY than for ND. When January–June precipitation was used, the tool correctly predicted 83% of the years with FGP reduced by drought for CO, 82% for WY, and only 67% for ND. Positive values of the True Skill Statistic (0.53 for CO, 0.42 for WY, and 0.17 for ND) indicate that FGP was classified as above or below average better than random selection. Predicting FGP earlier than April in CO and WY will require accurate forecasts of April–June precipitation. Use of the DC is most limited by insufficient forage data to determine the site-specific relationships between FGP and monthly precipitation. Even so, the decision tool is useful for discriminating drought effects on FGP classification being above or below the long-term average, and it provides a quantitative prediction to producers for their destocking decisions in drought years.  相似文献

A monoclonal antibody was raised with specificity for the granule bound starch synthase (GBSS1) Wx-B1 homeoallele of wheat (Triticum aestivum L.) using a synthetic peptide immunogen. This was used to develop a simplified enzyme-linked immunosorbent assay (ELISA) for the discrimination of Wx-B1a and Wx-B1b alleles which differ in the starch properties they confer. Discrimination of these alleles is important for the selection of wheat lines for Udon-style noodle production. The simplified ELISA worked in a single antibody (indirect) format and gave improved ease of use, discrimination of alleles and resolution relative to a previously developed 2-antibody (sandwich) ELISA. When the test was validated using breeders seed of a panel of commercial cultivars, heterogeneity of Wx-B1 alleles was observed for a significant proportion of the cultivars tested and this was confirmed using PCR analysis of the Wx-B1 and Wx-A1 genes. This observation has implications for cultivar wheat quality assessment, the application of molecular markers for variety identification purposes and the establishment of mapping populations from commercial cultivars.  相似文献
